Kisspeptin & Upstream Control of the HPG Axis

KNDy Neurons, Neurokinin B, Dynorphin, and the GnRH Pulse Generator

Evidence grade 3 of 5, Mixed. Read how the Atlas grades evidence.

FDA
Investigational (research peptide; not approved for low testosterone)

What can go wrong

Kisspeptin is investigational. Research-grade or gray-market peptides marketed for libido or testosterone are not quality-controlled, not proven effective, and not approved. Manipulating the pulse generator without understanding can suppress rather than stimulate the axis.

This is something to take to a clinician. The Atlas can tell you what is known about kisspeptin & upstream control of the hpg axis in general. It cannot tell you what is happening in your body, and the difference matters most for exactly the symptoms that bring people to this page.

Kisspeptin, encoded by KISS1 and acting via the KISS1R/GPR54 receptor, is the principal upstream activator of GnRH neurons. Hypothalamic KNDy neurons (kisspeptin, neurokinin B, dynorphin) in the arcuate nucleus are now considered the core of the GnRH pulse generator. Inactivating mutations in KISS1 or KISS1R cause hypogonadotropic hypogonadism and absent puberty; kisspeptin is under active investigation as a diagnostic and therapeutic tool.

The master switch sitting one level above GnRH, the true gatekeeper of the male axis.

What people take it for

These are the reasons people give for taking it. They are not effects it has been shown to have.

  • Explaining congenital hypogonadotropic hypogonadism
  • Research probe of GnRH pulse generation and fertility
  • Investigational stimulation testing of the reproductive axis

How it is supposed to work

Kisspeptin neurons sense steroid feedback and metabolic signals (including leptin) and drive pulsatile GnRH release. Within the arcuate nucleus, neurokinin B starts and dynorphin stops each pulse, while kisspeptin relays the signal to GnRH neurons. This KNDy circuit sets the frequency that ultimately determines LH, FSH, and testosterone output.

Kisspeptins are a family of peptides cleaved from the KISS1 precursor (kisspeptin-54 and shorter fragments) that share a common C-terminal that binds KISS1R.

Why kisspeptin is not a shortcut to higher testosterone

Because kisspeptin acts above GnRH, sustained administration can desensitize the axis just as continuous GnRH does, rather than reliably raising testosterone. Its likely clinical value is diagnostic (probing axis integrity) and in fertility, not as a self-administered testosterone booster. Gray-market 'kisspeptin' peptides are unapproved and unproven for that purpose.

What it does to fertility

Pulsatile kisspeptin (like pulsatile GnRH) can in principle stimulate gonadotropins and is being studied to induce or assess fertility; continuous exposure may instead suppress it. It is not an established fertility treatment.

What people get wrong

  • Often saidA kisspeptin nasal spray will boost your testosterone

ActuallyUnproven. Kisspeptin is investigational, and continuous dosing can desensitize the axis; marketed peptides are unregulated.

Who profits, and who pushes back

Nobody is named here yet, because we have not done that work on this entry. Read that as unfinished. Somebody almost certainly sells this, and somebody almost certainly disputes it. Assume both until we have checked.

What this entry does not yet cover

What follows is unfinished work on our side, not a finding about Kisspeptin & Upstream Control of the HPG Axis. A short entry is not a safe entry.
