Spermidine
Spermidine / Polyamine (Wheat Germ, Soy, Aged Cheese)
Evidence grade 2 of 5, Thin. Read how the Atlas grades evidence.
Evidence sits at preliminary/observational. The mechanistic story is genuinely strong in model organisms: Eisenberg's Madeo-lab work (Nature Cell Biology 2009; Nature Medicine 2016) showed spermidine induces autophagy and extends lifespan in yeast, worms, flies, and mice, and is cardioprotective in aged and hypertensive rodents. Human support rests on dietary-intake epidemiology, chiefly the Bruneck cohort (Kiechl, Am J Clin Nutr 2018), where higher dietary spermidine was associated with roughly 26 percent lower all-cause mortality. But the one adequately powered human RCT of a supplement, the SmartAge phase 2b trial (Schwarz, JAMA Network Open 2022; 100 adults, 0.9 mg/day wheat-germ spermidine, 12 months), was null on its primary memory-discrimination endpoint (treatment effect -0.03; P=.47), despite an encouraging earlier 30-person pilot (Wirth 2018). Autophagy induction in humans at supplemental doses remains unconfirmed. A correctly labeled 2 reflects that the causal human clinical case is not yet made.

What can go wrong
Well tolerated at studied low doses, but the human clinical benefit is unproven (primary RCT was null), and a theoretical cancer-proliferation caution plus a musculoskeletal-event signal warrant clinician input for at-risk groups.
Spermidine is a naturally occurring polyamine found in the body and, importantly, in food: wheat germ, soy, aged cheese, mushrooms, and legumes carry meaningful amounts. It draws its name from Antonvan Leeuwenhoek's 1678 microscopy of seminal fluid, though its scientific interest today comes from a different place. The Madeo laboratory in Graz showed that spermidine switches on autophagy, the cell's recycling and cleanup process, and extends lifespan across yeast, worms, flies, and mice. That preclinical record is unusually consistent. The human evidence is thinner and more mixed. Dietary-intake studies associate spermidine-rich eating with lower mortality and less cognitive decline, but the best controlled test in people, the SmartAge trial, found no memory benefit from a wheat-germ supplement over a year.
The autophagy-inducing polyamine, substantial longevity research framework.
What people take it for
These are the reasons people give for taking it. They are not effects it has been shown to have.
How it is supposed to work
Spermidine is a naturally-occurring polyamine, a small organic molecule (chemically N-(3-aminopropyl)-1,4-diaminobutane) found across virtually all living cells from bacteria through humans, where it functions as a foundational regulator of cellular metabolism, gene expression, protein synthesis, and substantially the induction of autophagy (the cellular self-cleaning process by which cells degrade and recycle damaged organelles, misfolded proteins, and metabolic byproducts). The substantial autophagy induction framework is the primary pharmacological positioning for spermidine supplementation. Endogenous spermidine biosynthesis declines substantially with age across most tissues; dietary spermidine intake (from wheat germ, soybeans, mushrooms, aged cheeses, natto, mature plant foods, and various other dietary sources) provides substantial supplementation framework. The substantial Eisenberg 2009 Nature Cell Biology foundational paper from Frank Madeo's laboratory at the University of Graz demonstrated substantial spermidine-induced autophagy across yeast (substantial lifespan extension), Drosophila (substantial lifespan extension), C. elegans (substantial lifespan extension), and human peripheral blood mononuclear cells, establishing the substantial polyamine-autophagy framework that has shaped subsequent research through the 2010s-2020s. The substantial pharmacological mechanism centers on EP300 acetyltransferase inhibition. Spermidine inhibits the EP300 histone acetyltransferase, which functions as a substantial autophagy suppressor through acetylation of multiple autophagy-related proteins (ATG5, ATG7, LC3, others). The substantial EP300 inhibition by spermidine de-suppresses autophagy machinery, allowing substantial autophagosome formation, lysosomal degradation of damaged cellular components, and broader cellular cleanup. The substantial autophagy induction framework provides theoretical mechanistic foundation for substantial spermidine-induced lifespan extension across model organisms, autophagy is among the substantial conserved longevity mechanisms across species (caloric restriction induces autophagy; mTOR inhibition induces autophagy; rapamycin induces autophagy; intermittent fasting induces autophagy). The substantial spermidine-autophagy framework has substantial mechanistic plausibility for human longevity applications, though substantial human clinical evidence remains limited. Secondary mechanisms include substantial mitochondrial biogenesis support (spermidine modestly induces PGC-1α and substantial mitochondrial protein synthesis), substantial anti-inflammatory effects (modest NF-κB modulation, modest TNF-α and IL-6 reduction in some preclinical models), substantial cardioprotective effects (modest endothelial function support, modest left ventricular function preservation in aging mouse models, Eisenberg 2016 Nature Medicine), substantial neuroprotective effects (modest preclinical Alzheimer's and Parkinson's disease model evidence through autophagy induction supporting amyloid-beta and alpha-synuclein clearance), and substantial hair follicle support (preliminary evidence for hair shaft elongation in cultured human follicles). The substantial epidemiological evidence framework provides substantial population-level support distinct from clinical trial framework. The substantial Bruneck Study (Italian-Austrian Tyrol cohort, Kiechl 2018) demonstrated substantial inverse association between dietary spermidine intake (estimated from food frequency questionnaires) and all-cause mortality across approximately 800 adults followed for substantial follow-up periods, substantial mortality risk reduction in highest-quartile spermidine intake versus lowest-quartile, comparable to other substantial dietary intervention effect sizes (Mediterranean diet adherence, etc.). The substantial Eisenberg 2018 NEJM analysis of similar cohort data provided substantial mortality and cardiovascular outcome association support. The substantial epidemiological framework provides substantial population-level support though substantial confounding variable concerns (diets high in spermidine are also high in substantial other nutrients) and substantial reverse causation concerns warrant interpretation calibration. The substantial pharmacokinetic profile reflects substantial absorption complexity. Oral spermidine has substantial dose-dependent absorption with substantial gut bacterial metabolism creating substantial individual variability in systemic exposure. Approximately 30-60% of oral spermidine appears to reach systemic circulation in pharmacokinetic studies; substantial variation across individuals based on gut microbiome composition. The substantial gut microbial polyamine production (predominantly E. coli and various other commensal bacteria produce substantial endogenous spermidine) creates substantial systemic exposure independent of dietary intake. Wheat germ extract supplements (the substantial commercial spermidine supplement format) provide approximately 1-2mg spermidine per serving; substantial dietary intake from food sources can reach 10-25mg daily in spermidine-rich diets (substantial natto, aged cheese, mushroom, wheat germ consumption). The substantial commercial supplement category has expanded substantially through the 2010s-2020s. spermidineLIFE (Longevity Labs/TLL Healthcare, Austrian-based, substantial Madeo lab institutional connection through Graz University spinoff) is among the substantial premium spermidine supplement brands with substantial wheat germ extract sourcing and substantial premium pricing positioning. Various other brands (Double Wood, Life Extension, ProHealth Longevity, others) provide substantial commercial alternatives at substantial price-point variation. The substantial Bryan Johnson Blueprint protocol inclusion (1mg spermidine daily through wheat germ extract supplementation) reflects substantial premium consumer longevity protocol category positioning. The substantial pharmaceutical category remains essentially absent, spermidine is positioned exclusively as dietary supplement framework without substantial pharmaceutical category development through 2025.
How it is taken
Spermidine supplementation is typically dosed at 1-2mg daily through wheat germ extract, the substantially most-studied supplementation format based on substantial Madeo Lab Graz University research framework. The substantial spermidineLIFE premium positioning, Double Wood mid-tier positioning, Life Extension and ProHealth Longevity mainstream positioning, and Bryan Johnson Blueprint protocol inclusion all use substantial wheat germ extract format at substantially similar dosing ranges. The substantial supplement category dosing reflects substantial calibration to research framework rather than substantial therapeutic dose-response characterization, human clinical trial dose-response evidence remains limited. Take with or without food, wheat germ extract absorption is reasonable either way. Many users prefer morning dosing as part of routine longevity protocol integration. Substantial sustained daily supplementation matters more than acute dosing, autophagy induction operates through sustained cellular metabolic signaling rather than acute pharmacological effect. Substantial supplementation duration matters, meaningful autophagy modulation likely requires substantial sustained supplementation over months-to-years; substantial epidemiological mortality association developed across substantial follow-up periods (Bruneck Study substantial follow-up across years). Dietary spermidine intake provides substantial supplementation alternative or complement. Substantial dietary spermidine through wheat germ (~13mg per 100g), soybeans (~10mg per 100g), aged cheese (~3-9mg per 100g varies by aging), mushrooms (~3-9mg per 100g varies by species), natto (~14mg per 100g), green peas (~4mg per 100g), and various other sources can substantially exceed typical wheat germ extract supplement dosing (1-2mg). Substantial spermidine-rich diet can reach 10-25mg daily, substantial Mediterranean diet adherence with substantial mushroom, aged cheese, and whole grain consumption provides substantial dietary supplementation framework. The drug interaction profile is modest for most users. Substantial gut microbiome polyamine production creates substantial individual variability in systemic spermidine exposure; substantial antibiotic disruption may modestly affect endogenous polyamine production. The substantial cancer-polyamine interaction creates substantial coordination context for users with active cancer or recent cancer history, polyamines including spermidine are substantially upregulated in many cancer cell metabolisms; specialist oncology coordination essential. The substantial wheat germ allergen profile creates substantial coordination context for users with celiac disease or wheat allergy. Pregnancy and lactation: limited safety data; specialist OB-GYN coordination reasonable. Pediatric use: not appropriate without specialist coordination. The substantial premium positioning reflects substantial Madeo Lab Graz University research framework inheritance; substantial mainstream alternatives provide substantial accessibility at substantial cost reduction. The substantial Bryan Johnson Blueprint protocol inclusion reflects substantial premium consumer longevity protocol category positioning. The substantial dietary positioning advantage (whole foods nutritional matrix versus isolated supplementation) suggests substantial dietary approach as foundational supplementation framework with supplements as substantial supplement.

What people get wrong
Often saidSpermidine is a "miracle longevity supplement" that will extend my lifespan.
ActuallySubstantial preclinical evidence supports substantial lifespan extension in yeast, Drosophila, C. elegans, and mice. Substantial human translation evidence remains limited, substantial mechanistic plausibility with substantial clinical outcomes uncertainty. Reasonable substantial supplementation framework with substantial mechanistic positioning; calibrated expectations regarding substantial human lifespan outcomes.
Often saidI should take very high doses to maximize autophagy benefits.
ActuallyStandard supplementation doses (1-2mg daily through wheat germ extract) reflect substantial research framework calibration. Substantial dietary intake (10-25mg daily through spermidine-rich diet) substantially exceeds typical supplement dosing without substantial safety concern. Substantial autophagy induction operates through sustained cellular metabolic signaling rather than acute high-dose pharmacological effect.
Often saidWheat germ extract is the only effective spermidine source.
ActuallySubstantial dietary spermidine through whole foods (wheat germ, soybeans, aged cheese, mushrooms, natto, green peas) provides substantial supplementation framework with substantial nutritional matrix advantage. Substantial dietary positioning advantage through cofactor polyphenol contributions and broader nutrient framework. Reasonable dietary approach as foundational supplementation framework with supplements as substantial supplement.
Often saidSpermidine supplementation is dangerous because polyamines are linked to cancer.
ActuallySubstantial cancer-polyamine interaction creates substantial coordination context for users with active or recent cancer history, specialist oncology consultation essential. Substantial general adult supplementation framework remains reasonable for healthy adults without substantial cancer history. Substantial individual risk-benefit assessment essential rather than blanket avoidance.
Often saidEndogenous spermidine production declines with age, so supplementation is essential.
ActuallySubstantial endogenous polyamine biosynthesis does decline modestly with age across most tissues; substantial dietary spermidine intake can substantially compensate. Substantial supplementation framework is reasonable but not strictly essential, substantial dietary patterns (Mediterranean, traditional Japanese) provide substantial polyamine intake without supplementation.
Often saidspermidineLIFE is medically superior to other spermidine supplements.
ActuallyspermidineLIFE has substantial Madeo Lab institutional connection through Graz University spinoff providing substantial research framework inheritance; substantial premium positioning. Substantial mainstream alternatives (Double Wood, Life Extension, ProHealth Longevity) provide substantial similar wheat germ extract format at substantially lower price point. Reasonable consumer choice based on price/positioning preferences rather than substantial medical superiority claims.
Often saidSpermidine is a "natural" so it has no safety concerns.
ActuallySubstantial endogenous production and substantial dietary intake establish substantial general safety profile. Substantial coordination considerations remain, cancer history, wheat allergy/celiac, pregnancy/lactation, pediatric use, autophagy-related rare diseases all warrant specialist coordination.
Who profits, and who pushes back
Limited human clinical trial evidence
Despite substantial preclinical evidence (yeast, Drosophila, C. elegans, mouse model lifespan extension) and substantial epidemiological evidence (Bruneck Study mortality association), substantial human clinical trial evidence for substantial longevity outcomes remains limited. The Schwarz 2018 human pilot demonstrated autophagy marker increases; the SmartAge 2021 cognitive trial did not demonstrate substantial cognitive benefit. Mainstream geriatrics and longevity research scrutiny calibrates the substantial evidence base toward substantial mechanistic plausibility with substantial translation uncertainty. Reasonable position: spermidine supplementation has substantial mechanistic foundation and substantial epidemiological support; substantial clinical trial evidence remains aspirational; reasonable supplementation framework with substantial mechanistic positioning rather than substantial established clinical outcomes.
Polyamine-cancer interaction complexity
Polyamines including spermidine are substantially upregulated in many cancer cell types; substantial cancer cell growth dependency on polyamine metabolism in many cancers. The substantial DFMO/eflornithine pharmaceutical category targets polyamine biosynthesis inhibition for some cancer applications, substantial pharmaceutical evidence supporting polyamine reduction for cancer treatment. The substantial polyamine-cancer interaction creates substantial complexity for users with active or recent cancer history considering spermidine supplementation. Mainstream oncology calibrates this consideration toward substantial coordination essential. Reasonable position: spermidine supplementation requires substantial oncology coordination for active or recent cancer history; substantial general adult supplementation framework remains reasonable for healthy adults without substantial cancer history; substantial individual risk-benefit assessment essential.

The market for this
What follows describes how this is bought and sold. It is not a buying guide and it is not an endorsement of any seller.
Spermidine wheat germ extract sourcing involves substantial agricultural supply chains predominantly through European (Austrian, German, Italian, Polish) and Asian (Japanese, Chinese) wheat germ supply infrastructure. Substantial wheat germ extraction processes use ethanol or aqueous solvent extraction with substantial concentration of spermidine and broader polyamine content (substantial spermine, putrescine contributions in wheat germ extract). Substantial standardization to spermidine content (typically 0.8-1.2mg per supplement serving) reflects substantial quality control investment. The substantial spermidineLIFE premium positioning reflects substantial Madeo Lab Graz University institutional connection, substantial research framework inheritance translates to substantial premium commercial positioning. Substantial mid-tier alternatives (Double Wood, Life Extension, ProHealth Longevity) use substantial similar wheat germ extract sourcing through various commercial suppliers. The substantial Japanese natto-derived polyamine sourcing represents substantial alternative format for users seeking non-wheat sourcing, substantial bacterial fermentation production of polyamines through traditional natto preparation. Substantial fermentation-derived spermidine production reflects substantial emerging commercial category. Heavy metal contamination is not substantial concern for wheat germ extract sourcing from reputable suppliers; substantial quality control across mainstream supplement manufacturers addresses this consideration. Substantial allergen disclosure essential for wheat-derived supplements, substantial consumer coordination for celiac disease and wheat allergy users. The substantial dietary spermidine alternative through whole foods (wheat germ, soybeans, aged cheese, mushrooms, natto, green peas) provides substantial supplementation framework with substantial nutritional matrix advantage. The substantial European regulatory framework (EFSA, EU food supplement regulations) provides substantial regulatory inheritance for substantial European supplement category positioning. Substantial Madeo Lab Graz University research framework provides substantial pharmacological credibility through substantial Eisenberg 2009 Nature Cell Biology foundational paper and substantial subsequent research framework development.
