P21
NAPVSIPQ-derived Neurogenic Peptide
Evidence grade 1 of 5, Untested. Read how the Atlas grades evidence.
Evidence for P021 (the CNTF-derived neurogenic peptidergic compound Ac-DGGLAG-NH2) is entirely preclinical. The supporting work is animal and in-vitro only, chiefly from the Iqbal laboratory in 3xTg-AD and Down-syndrome mouse models and aged Fisher rats (Kazim 2014, Baazaoui/Iqbal reviews, CDKL5 in-vitro/in-vivo model 2024), reporting enhanced hippocampal neurogenesis, increased BDNF transcription, and reduced tau pathology. No human clinical trial has been registered or published, so on the honest scale this is animal-only and rates grade 1.
- FDA
- Not FDA approved. No IND-stage registered human clinical trial identified; preclinical (animal and in-vitro) only. Sold as a research chemical labeled not for human consumption.
- WADA
- Not listed by name on the WADA Prohibited List; however, as a growth-factor / neurotrophic-signaling agent it could fall under WADA S2 (peptide hormones, growth factors, related substances and mimetics) by class, and its unapproved status means it also carries S0 (non-approved substance) exposure. Athletes should not assume it is permitted.
Reference only. Not medical advice.
What can go wrong
Very limited data and no human safety testing; an unapproved research chemical with animal-only efficacy signals and unknown human dosing, purity, and long-term risk.
Talk to a clinician before taking this. The Atlas can tell you what has been studied about P21. It cannot tell you how it would interact with your body, your history, or anything else you are taking, and nothing on this page is a reason to start.
A peptide derived from the CNTF/NAP family with proposed BDNF-mimetic and neurogenesis-promoting properties. Early research; growing nootropic community interest for neuroplasticity.
What people take it for
These are the reasons people give for taking it. They are not effects it has been shown to have.
How it is supposed to work
P21 (also written P021) is a small peptide derived from a fragment of the neurotrophic factor CNTF, engineered to be drug-like and to cross into the brain. It promotes the birth of new neurons (neurogenesis) and boosts BDNF, and has been studied mainly as a potential treatment for Alzheimer’s disease and cognitive decline.
P21, also written P021, is a small peptide reverse-engineered from the most active region of ciliary neurotrophic factor, with an adamantane group added to help it survive in the body and cross into the brain, so it can be taken orally. It should not be confused with the unrelated p21 tumor-suppressor protein of cancer biology.

A CNTF Mimic For Memory, Promising In Mice From One Lab
P21 is a thoughtfully designed cognitive peptide whose honest weakness is the narrowness of its evidence base. The design solves a real problem: natural growth factors like CNTF and BDNF have powerful effects on the brain but poor drug properties and side effects, so P21 was built to capture CNTF’s benefit in a small, brain-penetrant, orally available molecule. Its proposed mechanism is coherent: it raises BDNF, apparently by blocking a signaling pathway called LIF, and higher BDNF dampens an enzyme that drives the abnormal tau tangling of Alzheimer’s, while also promoting new neuron formation and synaptic plasticity. The mouse results are genuinely striking: in an Alzheimer’s model, chronic oral dosing improved memory, increased BDNF, boosted neurogenesis and reduced tau pathology, with similar promise in a Down syndrome model. Two honest caveats dominate. The entire evidence base is preclinical and concentrated in the work of essentially one research group, which is a real reproducibility limitation, and the clean single-pathway story oversimplifies a still-incomplete mechanistic picture. There are no human trials, no approval and no established dosing.
Safety in detail
No human clinical trial has evaluated P021 for safety or efficacy. All positive findings are from rodent and cell-culture models in the Iqbal-lab research program. Vendor material is labeled for laboratory research use only and not for human consumption, and no regulator has cleared it for any human indication.
- P21 is a preclinical research peptide with no established human safety data.
- Its cognitive benefits are shown in animals, not proven in people.
- Gray-market versions, sold as nootropics, are unverified.
- Not approved for any use.
Children and teenagers
P21 has no validated pediatric use and is an unapproved research peptide affecting brain development pathways. There is no appropriate pediatric use.
What people get wrong
Nothing recorded here yet. There is almost certainly something people get wrong about this one, and the blank space means we have not written it down, not that the record is clean.
Who profits, and who pushes back
Nobody is named here yet, because we have not done that work on this entry. Read that as unfinished. Somebody almost certainly sells this, and somebody almost certainly disputes it. Assume both until we have checked.
P21 reaches users through the gray-market research-peptide and nootropic trade, sold for memory and cognitive enhancement on its mouse data. Its legitimate future would be as an Alzheimer’s drug candidate requiring human trials, which have not occurred.
How we got here
P021 was designed by Khalid Iqbal and colleagues (Nathan S. Kline Institute / NYU) as a synthetic mimetic of a short active fragment of ciliary neurotrophic factor, engineered with an adamantane-based amino acid for lipophilicity and blood-brain-barrier penetration and a C-terminal amide for peptidase resistance. It has been characterized in the peer-reviewed literature since roughly 2014, carries the development code GLXC-21260, and the underlying neurotrophic-peptide chemistry is covered by US patents assigned to that group, with reported development interest from Phanes Biotech (Pennsylvania) toward Alzheimer's disease. It has never entered registered human clinical trials and is instead distributed by research-chemical vendors (VPeptide, Core Peptides, Nationwide Peptides and others) under the names P21 and P021 for laboratory use only. The name is a persistent source of confusion because dropping the zero yields P21, which collides with the well-known cell-cycle protein p21 (CDKN1A); the compound in this entry is unrelated to that protein.
P21 was developed by a New York research group that reverse-engineered the active region of CNTF into a small, brain-penetrant, orally available peptide. Across mouse models of Alzheimer’s, Down syndrome and aging it improved memory, boosted neurogenesis and reduced tau pathology, while the evidence remained concentrated in that single group and untested in humans.

The people taking it
A neurogenic, CNTF-derived research peptide of interest in Alzheimer’s research, with an emerging gray-market nootropic following but no human proof.
P21’s users are nootropic and longevity enthusiasts seeking cognitive enhancement or Alzheimer’s prevention, plus the researchers pursuing CNTF-mimetic therapy for neurodegeneration.
Studied in research for neurogenesis and Alzheimer’s disease and used gray-market by some as a nootropic; it has no approved consumer use.
Set against the alternatives
P21 sits among the cognitive and neurotrophic peptides, near the memory peptide Dihexa, the neurotrophic mixture Cerebrolysin, the nootropic Semax and the neuroprotective NAP. Like Dihexa and the longevity peptide Epithalon, its main limitation is single-lineage evidence, a striking story resting heavily on one group’s work. Its distinctive strength is being orally available and brain-penetrant by design, unusual among peptides.
What we do not know
- Whether its neurogenic effects translate from animals to humans.
- Whether it can meaningfully affect Alzheimer’s disease.
- What its safety profile is, currently uncharacterized.
Legal status
P21 is a preclinical research peptide studied for neurogenesis and Alzheimer’s; it is not approved or marketed. It has some gray-market presence as a nootropic, where it is research-use-only material.
Status current as of July 2026. Regulatory positions change, sometimes quickly.
The market for this
What follows describes how this is bought and sold. It is not a buying guide and it is not an endorsement of any seller.
There is no approved consumer route. It exists mainly as a research compound; given the limited human data, self-experimentation is not advisable.
Not produced as an approved drug for this use; any consumer-facing material is research-use-only, typically synthesized overseas in China, with no quality assurance and frequently no certificate of analysis.
What this entry does not yet cover
